The mouse monoclonal antibody clone HI30 binds to all isoforms of human CD45 antigen (Leukocyte Common Antigen); CD45 is a 180-220 kDa single-chain type I transmembrane protein expressed at a high level on all cells of hematopoietic origin, except erythrocytes platelets and non-hematopoietic cells. It is a protein tyrosine phosphatase with several isoforms, each being generated and expressed in cell-specific patterns. With its broad cell distribution, CD45 is critical for many leukocyte functions, regulating signal transduction and cell activation associated with the T cell receptor, B cell receptor, and IL-2 receptor. Other forms of CD45, with restricted cellular expression, include CD45R (B220), CD45RA, CD45RB, and CD45RO. It is reported that clones HI30 and 2D1 anti-human CD45 clones can reciprocally block the binding of each other to CD45. For flow cytometric staining, the suggested use of this reagent is 5 μL per 1 million cells in a staining volume of 100 μL. If whole blood is analyzed, then use 5 μL per 100 μL. It is recommended that users titrate the antibody to obtain the optimal result for their specific application. Please briefly centrifuge the reagent vial before use.
